What is DNA Synthesis?

The definition of DNA synthesis is the process by which deoxynucleic acids combine to form DNA. Another name for DNA synthesis is DNA replication. DNA synthesis is the organic or artificial construction of deoxyribonucleic acid (DNA) molecules.

DNA is a macromolecule composed of nucleotide units linked by covalent and hydrogen bonds in a repeating structure. DNA synthesis occurs when these nucleotide units combine to form DNA. This process may be artificial (in vitro) or natural (in vivo).

DNA synthesis occurs in all eukaryotes, prokaryotes, and some viruses. Accurate DNA synthesis is necessary to avoid DNA mutations. In humans, DNA synthesis and the mechanisms involved in vivo have been extensively studied for decades, as mutations can lead to diseases such as cancer.

What are The Steps of DNA Synthesis?

DNA replication is when DNA creates a copy of itself in the cell division process. Here are the steps of DNA synthesis:

  1. Unravel the double helix system of the DNA molecule.
  2. An enzyme called a helicase helps this process, which breaks the hydrogen bonds that hold the complementary bases of DNA together.
  3. The separation of two individual strands of DNA creates a "Y" shape called the replication "fork". The two separated strands act as templates for building new DNA strands.
  4. One of the strands is oriented in the 3' to 5' direction (towards the replication fork) and is the leading strand. This process resulted in the replication of the two strands (leading strand and lagging strand)
  5. After matching the bases (A and T, C and G), an enzyme (exonuclease) removes the primers.
  6. The new strand is proofread to ensure the recent DNA sequence is error-free.
  7. Finally, an enzyme called DNA ligase seals the DNA sequence into two consecutive double strands.
  8. DNA replication results in two DNA molecules consisting of a new strand of nucleotides and an old strand of nucleotides.
  9. After replication, the new DNA automatically wraps around the double helix.

What is DNA Sequencing on DNA Synthesis?

DNA sequencing is part of DNA synthesis. DNA sequencing determines the order of bases (A, C, G, T) found in DNA.

Sanger sequencing is one of the earliest techniques that involves stopping DNA synthesis. This strategy was made possible by removing the hydroxyl group from the 3' end of the nucleotide.

When these 'dideoxynucleotides' are in the DNA strand, DNA polymerase cannot add the next nucleotide. The dideoxynucleotides are mixed with nucleotides to create fragments of different lengths.

By using dideoxy-A, dideoxy-C, dideoxy-G, and dideoxy-T in separate reactions, we can determine the final nucleotide of the part. After separating by size, we can decide on the DNA sequence by looking at the last nucleotide of the fragment by size.

Most modern DNA sequencing methods are automated and less labor-intensive than Sanger sequencing, although some took on Sanger's base of strategy. For example, we can add different fluorescent dyes to each dideoxy-nucleotide and detect differences by different fluorescent colors.

What are The Requirements for DNA Synthesis?

Watson & Crick's discovery of the double-helical nature of DNA explains how we could replicate and pass the genetic information to the next generation. The strands of the double helix can be separated and serve as templates for the synthesis of daughter strands.

In conservative replication, two daughter strands are transferred to her one daughter cell, and two parental strands go to the other daughter cell. In a semi-conservative synthesis process, one parent and one daughter strand would move to each daughter cell. Dr Noell Sturm explained the four components of DNA synthesis:

  • Substrates: The DNA synthesis process needs four deoxyribonucleotide triphosphates (dNTPs). The difference between deoxyribonucleotides and ribonucleotides is the lack of the OH group at the 2' position of the ribose ring.
  • Template: Nucleotides are incorporated into the growing DNA strand by base pairing with the template strand of DNA. A template is a DNA that copies onto a complementary strand of DNA.
  • Primer: DNA polymerase, the enzyme DNA, can only add nucleotides to an existing strand of DNA or RNA or primer base-paired to the template.
  • Enzymes: DNA polymerase should attach the incoming nucleotide to the primer. In initiating DNA replication, many other proteins and enzymes should assemble into large complexes called replisomes. DNA is thought to be spooled via the replisome and replicated as it passes through.
